Tubulin is not Posttranslationally Modified (Acetylated or Polyglutamylated) in Tetrahymena Macronucleus
نویسندگان
چکیده
In the interphase of the cell cycle of Tetrahymena pyriformis GL tubulin was not demonstrated in the macronucleus by confocal microscopy using antibodies to α-tubulin, acetylated α-tubulin and polyglutamylated tubulin antibodies or Flutax-1. During cell division, from the formation of the new oral apparatus to the beginning of cytokinesis, tubulin was diffusely present in the nucleus, however (i) it did not form visible microtubules in confocal microscopeand (ii) it was not acetylated or polyglutamylated. As acetylation stabilizes microtubules, the results point to the dynamism of nuclear tubulin which could have a role in the transport of the numerous macronuclear chromosomes.

Acetylation of lysine 40 in alpha-tubulin is not essential in Tetrahymena thermophila
In Tetrahymena, at least 17 distinct microtubule structures are assembled from a single primary sequence type of alpha- and beta-tubulin heterodimer, precluding distinctions among microtubular systems based on tubulin primary sequence isotypes. Tetrahymena tubulins also are modified by several types of posttranslational reactions including acetylation of alpha-tubulin at lysine 40, a modificati...

متن کاملPosttranslational modifications of alpha tubulin: detyrosination and acetylation differentiate populations of interphase microtubules in cultured cells
Subsets of microtubules enriched in posttranslationally detyrosinated (Gundersen, G. G., M. H. Kalnoski, and J. C. Bulinski. 1984. Cell. 38:779) or acetylated (Piperno, G., M. Le Dizet, and X. Chang. 1987. J. Cell Biol. 104:298), alpha tubulin have previously been described in interphase cultured cells. In this study an immunofluorescence comparison of these minor populations of microtubules re...
متن کاملDistinct Functional Roles of β-Tubulin Isotypes in Microtubule Arrays of Tetrahymena thermophila, a Model Single-Celled Organism
BACKGROUND The multi-tubulin hypothesis proposes that each tubulin isotype performs a unique role, or subset of roles, in the universe of microtubule function(s). To test this hypothesis, we are investigating the functions of the recently discovered, noncanonical β-like tubulins (BLTs) of the ciliate, Tetrahymena thermophila. Tetrahymena forms 17 distinct microtubular structures whose assembly ...
متن کاملPosttranslational modification and microtubule stability
We have probed the relationship between tubulin posttranslational modification and microtubule stability, using a variation of the antibody-blocking technique. In human retinoblastoma cells we find that acetylated and detyrosinated microtubules represent congruent subsets of the cells' total microtubules. We also find that stable microtubules defined as those that had not undergone polymerizati...
